Quantifying the Resolution of a Leica SR GSD 3D Localization Microscopy System with 2D and 3D Nanorulers

DNA origami based nanorulers produced by GATTAquant are the perfect positive control samples that tell you in a few minutes the status of your microscope and help you to test settings and configurations of your super-resolution microscope. Recently the nanorulers were used to test the performance of the Leica SR GSD 3D microscope. We used two-dimensional as well as three-dimensional structures which were imaged super-resolved by performing the localization based super-resolution technique DNA-PAINT. The results demonstrate that these nanorulers are optimal structures for resolution quantification. Also it showed that the Leica SR GSD 3D system is a well performing super-resolution microscope for localization microscopy with a lateral resolution of better than 40 nm and an axial resolution of better than 80 nm.
